The largest and most cited trial, the NIH-funded GAIT study, found that glucosamine and chondroitin, alone or combined, did not significantly outperform placebo for the overall study population — though a subgroup with moderate-to-severe pain showed a possible benefit from the combination. This is the honest, complicated starting point for any claims about this duo.
Why the picture is still not simple
Since GAIT, other reviews and meta-analyses have found more favorable results, particularly for glucosamine sulfate specifically (as opposed to glucosamine hydrochloride, a different salt form) at doses of 1,500 mg/day. Form and dose both matter more than the ingredient name alone.
What doses research actually uses
